   I can confirm, wolf is not on the island and the quest doesn't pop up. OP should maybe rexamine the file or take another crack at it

   Considering that we can't access his inventory, is there any way to know how much food he have left? Assuming we gave him 20 or 30 units of meat for example.

      While I see what you mean... that seems a bit unrealistic. I mean, this is a wild animal that leaves if you don't feed it. Would such a creature honestly consent to carrying your burdens and wearing dog armour?
