Have used Loot for the load order, but no luck. Normally, my game loads in 18 - 30 seconds, but with this mod active, it takes 2 - 3 minutes and as soon as the game starts I get CTD.
First of all you have to download and install correctly SKSE. Secondly, you have to download and install correctly SkyUI. Third, run the game via SKSE exe file. Four, to enable correctly mod you have to enter in "Mod Settings" menu (Esc -> MOD SETTINGS) and quit (Tab -> Tab). After all steps you will see in right top corner the message "Realistic Regeneration disabled".
Then you will be able to configure the mod settings (Esc -> MOD SETTINGS -> Realistic Regeneration).

P.S. Most of mods written for SKSE/SkyUI/MCM does not require any load order sorting. They uses functions provided from skse's dll file internally.
This mod changes some magic effects for spells (they are just buggy in vanilla game) and you have to place this mod at bottom (after described next mods) of load list only when you are using some mods for potions, food (for instance, Babbete's food mod).
P.S.II. If you dislike SkyUI interface you may try to install SkyUI Away mod which reset in-game user interface to vanilla one but you are able to use mods written for SKSE/SkyUI still.
