You don't have to wait BTW - enter vault, do Austin's tour, get the quest to find the cat that ran outside, give blood, go get the cat, and when you come back to the vault "Hole In The Wall" will be available. Has worked for me in every playthrough I've done it in.
I usually just (in console) disable the exit doorway door (the one in the vaults entry way) then enable it once i'm past it and unlock it proper using its linked terminal then just go down the elevator grab curie right there and back up, no molerat hassle, no need to pick between who gets cured and no bug of not being able to save in there past a certain point.
So if you wanted to make a mod that doesn't break a few things in the vault (or least it might not) just have that door unlocked from the start and recommend people not use it until the quest is started for better immersion (unless they just want to rush it) removing walls causes issues with visibility etc in the area and makes everything white and moving weird :)
If you clip through the exit (tcl console command) of the curie quest at the vault entrance and take the elevator down to curie, the kid will be already on the clinic bed ready for your antidote, without any pre-quests, and dont need to fight molerats too.
its the same with BOS reinforcement, if you clip through the exit of fort hagen and use the terminal, you will get Prydwen flying over.
