First! Setting up SkyrimSE with CPU Priority and Affinity, I gained roughly 10fps in heavy areas like Riften forrest and overhauled cities (Capital Windhelm/Whiterun). Keep in mind, results will vary based on your hardware and Skyrim mod list, but do see if you can try the below.
Second! I'll be documenting boring3 and oskkar's instructions with my own as viable options for cpu priority and affinity setup. Keep in mind, boring3's Skyrim Priority (and possibly DevourerPi's Skyrim Memory Optimizer - Plugins ) is planning to create/update their DLL's to include CPU affinity. So, keep a watch out for their mods.

These MO2 configuration instructions build off xrayy's instructions in his xrayys Optimized Skyrim CPU Balance mod. These instructions can also be used to create desktop shortcuts if you don't use MO2.
Thanks to Boring3 and oskkar, we actually have the steps to set CPU Priority and Affinity in MO2 or desktop shortcuts.
Please see the comments sections' sticky comment for their gracious steps. oskkar's solution is very clear and flexible in handling the cpu affinity.
Keep in mind Boring3's example has cpu affinity = FF5 which is only for a 12 cpu setup. You'll need to determine the appropriate hexadecimal value for your system and replace FF5 with yours.
Please use their steps instead of mine. See the sticky post in the comments. I'll keep my steps for future reference and humor.
