The Diamond City Greenhouse (next to Valentine's Detective Agency) contains one of the most advanced (but also, in retrospect, not really all that advanced) pieces of technology in the Commonwealth. Presumably by snooping on TEMPEST-type emanations from synth infiltrators, this bat is sensitive to their presence in your populous (> 4 person) settlements.
Just go and bash one of your settlers. If they are completely unharmed, there is not a synth spy about here. If they wince and react electrically, a synth is present at (or merely assigned to) this settlement. If their head blows off, well, you've discovered a dirty synth spy and will earn a nominal amount of XP for your trouble.
That's pretty much it.
Known Issues: If you have the Mr. Sandman perk, let sleeping synths (and innocent settlers) lie. The instant kill on sleeping persons park of that perk will override any non-hostile nature of weapons.
